Alkenone-inferred temperature is derived from obtained from the sediment of Lake Xiarinur in mid-latitude China.Freeze-dried samples were soxhlet extracted with dichloromethane: methanol (9:1 v/v) for 48 h. Dried total lipid extracts were redissolved in 1 mL 6% potassium hydroxide in methanol overnight at room temperature to remove interfering alkenonates. The n-hexane extracts were separated into subfractions using a 3 mL Alltech sol-id-phase extraction (SPE) tube containing 0.5 g of silica gel. The samples were then eluted with hexane (F1: hydrocarbons) and 1:1 dichloromethane-hexane (F2: alkenones). The F2 elution containing alkenones was evaporated under a nitrogen stream and redissolved in toluene. Prior to GC analyses, the extracts were derivatized overnight with bis (trimethylsilyl) tri-fluoroacetamide (BSTFA) at room temperature and then analyzed with a SHI-MADZU GC-2010 gas chromatograph, using nitrogen as the carrier gas. |
